Students, profs. debate effects of debt forgiveness | The Daily Free ...

While a report by the New America Foundation said the intended changes to President Barack Obama?s administration?s student debt-relief plan might not benefit all borrowers, a number of Boston University students and professors said they still believe the plan will be effective.

Borrowers pay 15 percent of their discretionary income and the remaining balance with accrued interest is forgiven after 25 years of payment. The Obama administration aims to change this policy to ask for 10 percent of the borrower?s discretionary income and forgive balances after 20 years.

However, Alex Holt, co-author of the report released Tuesday, ?Safety Net or Windfall?? said the Income-Based Repayment plan disproportionately helps some borrowers when he studied what happens to a borrower during the entire repayment period.

?If this new policy goes into effect, it will disproportionally help high-income borrowers, people who are making a lot of money,? Holt, a Federal Education Budget Project program associate, said. ?Our question is, ?Is that the best way to appropriate federal funds?? Should federal funds be going to the wealthiest earners in the country, or should it be going to borrowers who are struggling the most.?

The report offers recommendations to alter the policy to redistribute the relief to those who need it.

?We?re trying to take what we think is the intent of the law, which is to help struggling borrowers,? Holt said. ?We want to keep those policies in place to help those struggling borrowers, but we want to make sure people who can pay back their loans in full, do.?

Holt used the example of an attorney making $200,000 a year. After the attorney is out of school and working for 25 years, he or she might have $100,000 left to pay back. At that point, the debt is forgiven even though he or she could potentially pay it back, Holt said.

?It hurts the taxpayer because they are basically subsidizing wealthy people,? he said.

The Obama administration?s new plan will benefit anyone with a student loan, no matter the size of the loan, said School of Management Professor Mark Williams.

Williams said focusing on the size of the loan is missing the point of the financial relief plan.

?College students when they get out, they?re not going to be strapped,? Williams said. ?Instead of having a 15-percent requirement of their income going to pay the student loan, 5 percent more is going to be available for other things.?

The distribution is not as disproportionate as the report claims, he said. The high-income borrowers will pay more in taxes because they are making more and, in a sense, they will be paying back for that advantage.

Williams said the plan gives an incentive for more students to continue to graduate school.

?The positive is this is a greater incentive for students to want to go to graduate school,? he said. ?If they go to graduate school, they make more money. If they make more money, more debt to get to this higher income producing status will be extinguished.?

Cathie Jo Martin, a political science professor, said this distribution of wealth is not different from what often happens to lower-income classes.

?The use of the tax code to achieve social purposes in America has resulted in many benefits being distributed to the upper-income and middle-income classes,? Martin said.

She said the Obama administration is attempting to give lower-income students the opportunity to loan access.

?Obama is trying to reform the system of Pell grant so that more lower-income students can have access,? Martin said. ?I think he has expanded access.?

College of General Studies sophomore Arlene Garcia said the plan is beneficial as long as everyone gets an equal opportunity to have the same assistance.

?But I don?t think they should penalize people for earning more money,? Garcia said.

Garcia said it would not be fair for the government to ask high-income borrowers to pay their loans in entirety when others get their balances forgiven.

?It being ?forgiven? doesn?t really mean that it?s forgiven,? said Alex Keiser, an SMG junior. ?Somebody still has to pay the bill, which means then it?s just the taxpayers or someone else and, I kind of disagree with that.?

Keiser said people should be responsible for their own expenses, despite their financial background.

?We should be focusing on decreasing the prices of schools rather than trying to help people pay for them,? said Lejla Huskic, a College of Arts and Sciences sophomore. ?It?s putting a band-aid on the wound before disinfecting. [It] won?t do anything but trap the disease, and it?ll burst eventually.?

People who have the means should continue to pay their loan bill, she said.

?You should pay a larger percentage based on how much money you make,? Huskic said. ?This way, regardless of class, you?re able to pay what you can afford.?

Early Metaphysical Ideas

Early Metaphysical Ideas

Are our actions the result of circumstances or are we directed by external influences such as Cosmic vibrations? There are individuals who are masters and creators of their destinies for part of their lives, and innocent victims of fate at other times.

Some individuals believe it is the environment that has gradually molded our civilization and made mankind what it is. Others believe you may want to lay aside that belief and think again. There is an ancient system of self-mastery that contains fundamental principles that suggest you have complete control over your fate and that mankind has always had it.

These fundamental principles were recorded centuries ago and were evolved through observation over time. The principles were founded and perfected by test and application and reveal that all life and all existence within the universe are expressed in cycles.

It was determined that the cyclic periods are equivalent to certain measured rhythmic stresses and cosmic impulses . So, at a certain time of the day there is a cosmic impulse that is specific for reaping the benefits of a specific human activity. Go work out at the gym during the G period on Friday, for example.

Enthusiasts who believe in these principles might set a wedding ceremony to begin at 5:30 pm on a Thursday in order to reap all the positive influences for their union. They have become masters of their fate because they have timed their union to begin when the cosmos is most in favor of marriage.

According to the true believers the newlyweds will have the advantage over couples who marry without this knowledge. The newlyweds believe there are cosmic vibrations that have negative effects on couples who marry during a time of negative influences related to marriage.

Modern scientists have found that there is a definite rhythm to the human breathing process and heart action that closely resembles the more pronounced rhythms of cosmic energy. These findings suggest that these vibrations influence all living things on the planet ? including plants. The findings also suggest that the influences can be timed and measured.

The ancients who studied these influences were convinced that we can become master of our fate by designing our activities around the daily cycles and time periods suggested in these studies. These studies began with ?The order of The Rosicrucians.? Further information regarding The Rosicrucians is available online.

Microsoft Surface pre-orders in Canada, UK are not delayed (Update)

Microsoft Surface preorderers in Canada, UK report release dates pushed back to November 2nd

Microsoft's Surface for Windows RT appears to be flying off the virtual shelves, however it seems some of the customers who've pre-ordered will be waiting about a week longer than they thought. We've received several tips from readers in Canada and the UK indicating they've received emails (as seen above, check after the break for similar reports from Twitter) explaining that their tablets will be shipping on November 2nd, instead of October 26th as originally scheduled when they put their requests in days ago. There's no reason currently listed for the delay, however we have contacted Microsoft for further comment on the situation. Currently the Surface is listed as completely sold out on the UK and Canada Microsoft Store sites, and in the US all three variants are no longer promised for delivery on release date. It's been reported that Microsoft is building up to five million of the devices for launch this fall, hopefully they can all find their way to good homes sooner rather than later.

Update: A Microsoft spokesperson has responded, informing us that the emails sent out were incorrect, so "all customers should expect their devices by the original date given." It will be following up with customers to let them know their shipping date has not changed, and you can read the full statement for yourself after the break.

[Thanks to Jack, everyone who sent this in]

University of Arkansas Selected to Host Regional Flag Football ...

Students play flag football on the intramural fields.

October, 2012

The University of Arkansas was selected to host the National Intramural Recreational Sport Association Regional Flag Football Tournament on Oct. 26-28.

?This is the first time an extramural tournament of this magnitude has been hosted on the University of Arkansas campus,? said Bill Mock, associate director of intramural sports for University Recreation.

A NIRSA Regional Flag Football Tournament presents an opportunity for intramural teams from different schools across a region and the country to play other teams from other schools after their own campus championships.?

?Having the chance to host a regional tournament is an honor because it is an opportunity to show off our university and the UREC intramural program to so many students and staff from across the country,? said Seth Davis, graduate assistant for Intramural Sports.??It is also an opportunity to help develop our student staff and players into professionals while also allowing the U of A intramural program to become more recognized among other recreation programs across the country.?

Intramural student officials come from all over the region to officiate these games. There are eight regionals across the country overall.

Champions from each regional receive stipends to travel and participate in the National Flag Football Tournament at the University of Central Florida in Orlando, Fla.?The top student officials also get selected and receive stipends to work the national championships.

Top 9 Tips For Building A Good Mobile Website | Business 2 ...

For many people, mobile devices have become the primary way of accessing the Internet. In fact, in some parts of the world, mobile internet usage has surpassed desktop internet usage.

In short, your audiences are fast becoming mobile? but is your website mobile yet?

Grab a smartphone.
Key in your website or blog URL.
If what you see is jumbled or distorted, you are likely losing tons of traffic?and conversions.

You need a mobile-friendly website?now.
But what does ?mobile-friendly website? mean? Here are nine tips, based on our experiences at?MobStac?(where I work), to help you build a solid mobile website.
Optimizing news websites to mobile websites
1. Display key information only
The homepage of your mobile website should display key information only. Your mobile visitors are mostly on the move, and the information they?re most likely looking for is your address, phone number, and address. Include those on your homepage.
You can provide additional, more detailed information via a well-defined navigation structure. But limit the total number of pages on your mobile website. If you try to fit in all your website content into your mobile site, you are merely miniaturizing your website, not mobilizing it.

The mobile homepage of the New York Times, for example, has a clear layout that displays the most important headlines on top, and it displays other sections as users scroll down.

2. Make the page load in less than three seconds

The attention span of your mobile visitor is very limited, so your mobile website?s page load time is crucial.

How long is your visitor willing to wait for a single Web page to load on your mobile phone? Nearly 60% of Web users say they expect a website to load on their mobile phones in three seconds or less.

If your mobile site does not load within three seconds, you?ll be losing visitors. So, avoid heavy graphics that increase page load time. Similarly, load only the minimum set of JavaScript and CSS files.

For modern mobile browsers,?use HTML5 to benefit?from its rich feature set.

3. Keep it light

Your mobile website should be devoid of large, Flash-based video files? for two reasons:

  • Most smartphone browsers do not support Flash.
  • Flash and heavy image files increase the page load time considerably.

Instead, use simple images to improve the look of your mobile website. All images used on the desktop website should be set to resize automatically to fit varying mobile screen sizes.

Use white space efficiently to give your mobile site a clean, simple, and organized look. (See the New York Times image, above, for an example of the efficient use of white space.)

4. Stick to your design ?language?

Incorporate the same branding elements and design language used in the desktop website for your mobile site, too. The use of those design elements on your mobile site will increase brand recall for users who are already familiar with your brand or main site.

5. ?Touch? is the new ?click?

The number of touch-based smartphones and tablets is increasing by the day. So make your mobile website touch-friendly.

Use big buttons, check boxes, and pre-populated fields whenever possible to help your mobile visitors input information easily at the touch of their keypad. Minimize the number of text input fields.

6. Don?t forget feature phones

When building a mobile site, remember that you are not just creating it for iPhone and other smartphone users. Also keep in mind visitors who own feature phones.

Your mobile website should work flawlessly on devices with various screen sizes and on various mobile platforms and operating systems, including iOS, Android, Blackberry, Nokia, and even feature phones that use Opera browsers.

Facebook?s mobile site?is a good example of a site that detects visitors? mobile device automatically and renders a version compatible with the device.

7. Redirect your mobile visitors automatically

After going through the hardship of building a solid mobile website, don?t forget the most important step: redirecting your mobile visitors automatically to your mobile website. Create a subdomain of your main website, typically, m.yoursite.com or mobile.yoursite.com, which is where your mobile website resides.

8. Track your mobile visitors

Depending on your location and your industry vertical or niche, your mobile website traffic might have already surpassed your desktop website traffic, so keep a tab on your mobile website analytics.

If you have an analytics system in place, including Google Analytics, create a separate profile for your mobile website. Doing so will help you track your mobile traffic and desktop traffic independently to better understand how your visitors behave differently on your mobile website vs. your desktop website.

8. Monetize intelligently

When you create a good mobile website, your mobile traffic is sure to surge. Have a plan ready for monetizing your mobile traffic.

If you rely on advertising revenue, note that your desktop banner ads will be too big for your mobile website. Sign up for a mobile ad network like InMobi or Google AdSense for Content to display ads according to the mobile visitor?s choice.

Remember, too, that the ads displayed on a smartphone might not work for feature phones.

Several PAR members speaking at this year's Triple Play ...

Friday is the early bird deadline for the Triple Play REALTOR? Convention and Trade Expo, Dec. 4-6 in Atlantic City.

Registration is only $79 when you register online at the newly updated RealtorsTriplePlay.com.

A new addition to the program this year is a special track, Registration PLUS. For an additional fee of $39, this track provides members with a guaranteed seat in five specific CE courses. The courses are held in one room and a lounge and caf? are nearby to purchase beverages, snacks, breakfast and lunch.

Registrants for Triple Play 2012 have the opportunity to gain access to: the industry?s most prominent speakers; the latest products and services at the Trade Expo; networking and referral opportunities; and much more.

Several Pennsylvania Realtors? are speaking at this year?s convention on topics including mobile technology, online marketing, social media and effective writing.

Realtor? Peter Lamandre of Scranton will offer an introduction to the Apple family of products and how Realtors? can integrate Apple into their business with his session iPad, iPhone, iCloud, iDon?t Know. ?Let?s face it, we live in a mobile world and we are in the process of breaking free from the tethered life of the PC,? Lamandre said.

?Many agents buy technology because it is the ?next cool thing? or ?because everyone else does? but few truly utilize the technology to its limits and never fully realize the benefit of their investment,? he added. ?The goal with this session is to move the needle and help agents realize the full potential of the devices while helping them close deals.

Realtor? Anne Rubin from Elkins Park will offer her CE session How to Structure a Lease Purchase. ?Lease purchases are becoming a more popular tool for moving some of the oversupply of houses on the market today,? Rubin said. ?Many potential buyers could have the ability to purchase a house if given the opportunity to apply a portion of their monthly rental payments to an escrow account in order to accumulate monies for down payment and settlement costs.

Agents need to understand how to structure a lease purchase that will protect and benefit the clients they are representing,? she added. ?After taking the course, agents will understand how to present a lease purchase to their clients, what the benefits and downfalls are and how to structure the agreement with a separate lease.?

Realtor? Chris Beadling of Doylestown will offer a quick overview of mobile marketing for Realtors? in his session Moving Beyond QR: A Guide to Mobile Marketing. ?Many real estate professionals are just starting to see the value in using their phones to leverage the Internet and are unsure what technologies are out there,? said Beadling. ?My hope is people will hear about some tools they may have never known before and realize that mobile marketing is more than just scanning boxes. People will learn that there are tools available even easier to use than Q.R. which are free and offer much more in terms of agent utility and mobility.

Visit the site today for full details including an interactive session schedule, speaker bios and to register.

Jason Aldean makes surprise announcement in Boston

BOSTON (AP) ? Jason Aldean will be making history at one of baseball's most historic sites.

Aldean will be the first country music star to play a concert at Fenway Park, the iconic home of the Boston Red Sox. The July 13 appearance will be the initial stop on his first stadium tour. Aldean made the announcement Thursday in the shadow of the Green Monster with an assist from Red Sox President/CEO Larry Lucchino and members of the team who made a celebratory clip for the singer that played on Fenway's video board.

"Being the first country music artist to ever come here and headline a show is amazing," Aldean said. "It's going to be a fun night. Over the last several years, my career, especially the touring side of that, has been pretty amazing. It's been growing. So thank you guys for having us. I can't wait for July 13."

The Georgia native released his fifth album, "Night Train," this week. That title is an apt metaphor for his career, which has been steaming along on a steep trajectory. About the only thing he hadn't attained yet was a stadium tour, and he's crossing that goal off the list.

Red Sox David Ortiz, Dustin Pedroia and Jarrod Saltalamacchia welcomed Aldean in a video, and he walked out of the dugout to the sound of songs from his new album and signed the wall inside the Green Monster scoreboard.

Aldean is one of country's top draws, but the stadium tour will move him into rare company. Currently, only Taylor Swift and Kenny Chesney have that kind of drawing power. But the hard-rocking singer has earned the status, selling more than 1.9 million tickets on his yearlong "My Kinda Party" tour that wraps in Dallas on Oct. 27.

Aldean will be joined in Boston next July by Miranda Lambert, Jake Owen and Thomas Rhett. The area has proven to be receptive to country music. Chesney and Swift have sold out Gillette Stadium, home of the New England Patriots, several times and Aldean has drawn enthusiastic crowds at smaller venues.

The stadium tour isn't the only sign Aldean's career continues to pick up speed. The 35-year-old singer is up for three awards, including top honor entertainer of the year, at the Country Music Association Awards on Nov. 1. The first single from "Night Train," ''Take a Little Ride," had the highest selling digital debut for a solo male country artist and was the fastest rising No. 1 on the country song charts this year.

Analysis: Presidential tax plans could bruise municipal bond market

(Reuters) - Tax policies proposed by the two presidential candidates may differ in several ways, but both could roil the $3.7 trillion U.S. municipal bond market, popular with wealthy investors and vital for the financing needs of states, cities and other issuers of tax-exempt debt.

Demand for munis, which offer investment income free from federal taxation, may be reduced if the proposals floated by either incumbent Democrat Barack Obama or Republican challenger Mitt Romney were to take effect.

That would push borrowing costs up for all municipal issuers that rely on the market.

Major tax reform encompassing munis would be the first since 1986. As Washington grapples with trillions of dollars of debt, comprehensive changes to the tax code appear likely, regardless of who is in the White House.

That prospect has set off some alarms in the muni market.

"The two candidates are presenting very different and frankly very confusing tax proposals as they relate to municipal bond interest," said Chris Mauro, head of U.S. municipals strategy at RBC Capital Markets.

An election guide for credit investors published by Morgan Stanley last month warned that "both policy sets are likely negative for munis' tax value and a headwind for performance."

LOWER RATES, FEWER BREAKS UNDER ROMNEY

Romney would lower all income tax rates by 20 percent while balancing those cuts by ending some yet-to-be-identified tax breaks, which could include munis, for high earners.

"Bottom-line, it seems it is going to be very hard for him to achieve what he wants to achieve without putting most of the very large tax expenditures in play and munis are one of the top 10 tax expenditures," Mauro said.

A January report from the U.S. Congress' Joint Committee on Taxation estimated that tax expenditures from public purpose and other muni debt would total around $230 billion between 2011 and 2015.

The Republican candidate's intention to eliminate taxation on capital gains, dividends and interest income for taxpayers with adjusted gross incomes below $200,000 would likely reduce demand for tax-free debt and push yields up, according to a recent report by Cadmus Hicks, a Nuveen Asset Management managing director.

Those taxpayers accounted for $35 billion, or about half of the tax-exempt interest individuals reported to the Internal Revenue Service in 2010.

Romney's plan for the below-$200,000 earners would also put corporate and U.S. Treasury debt, which typically offer higher yields than municipal debt because of their taxable status, on an even playing field with tax-exempt munis by freeing all of it from taxation, Morgan Stanley noted.

Still, investors with incomes greater than $200,000, who accounted for $34.2 billion of tax-free interest two years ago, may have room to expand their holdings of muni debt without demanding inflated yields, according to Hicks.

TAX BREAKS CAPPED UNDER OBAMA

Obama intends to keep the Bush-era tax rates for families making less than $250,000. The tax rate for annual incomes over that amount would be increased and the value of the muni tax exemption would be limited to 28 percent.

Higher rates for the wealthy would make tax-free munis more attractive to those investors, but capping the muni tax exemption would curb enthusiasm for the debt. Romney has also floated the idea of a monetary cap on tax deductions, but it is not clear if munis would be included.

What worries traders and dealers in the muni market is that Obama's 28 percent cap would include existing debt in the secondary market, a move that would slash the value of outstanding bonds.

"Under the proposal as they presented it, there would be no grandfathering," RBC's Mauro said. "All bonds would be subject to this 28 percent cap. The whole market would have to readjust its pricing."

According to a recent paper issued by the National Association of Bond Lawyers, the cap would reduce the value of a $1,000 bond maturing in 10 years to less than $950 -- a more than 5 percent drop.

Mauro said the Obama administration has suggested that the 28 percent cap would not be "rock solid" and that it could be even lower if certain deficit reduction targets are not hit.

"That would be debilitating for the municipal market because there would be so much uncertainty about how you would price in that tax risk," Mauro said.

ENDING THE ALTERNATIVE MINIMUM TAX

Both Obama and Romney would end the alternative minimum tax, which was enacted to put a floor on the amount of taxes paid by the rich but is hitting an increasing percentage of taxpayers every year.

The AMT is applied to earnings from a small percentage of muni bonds sold by issuers such as airports and housing authorities that have substantial private-activity components in their deals.

Obama's plan would require households earning over $1 million a year to pay a minimum tax of 30 percent.

The uncertainty surrounding the different proposals and the prospect of drastic change has led many market participants to circle the wagons to protect tax exemption. For example, Municipal Bonds for America, a coalition representing dealers and issuers, was launched last week by the Bond Dealers of America to defend tax exemption.

Peter Hayes, head of BlackRock's municipal bonds group, said while the wealthy generally benefit from investing in munis, eliminating tax-free muni bonds would hit all taxpayers in the form of higher borrowing costs for governments.

But intense lobbying may stave off changes to tax exemption, particularly if the control of the federal government remains split between the political parties, he said.

"Because there is so much uncertainty and because the possibilities are so vast, it may actually in the end be hard to get anything done around tax policy or tax reform," Hayes said.

Acer reportedly looking to launch Intel powered smartphone before the year end

Android Central

It's inevitable that we'll start to see more Intel powered Android devices as we move into 2013. But, the latest word to reach us from the far east suggests we may yet see another before this year is out. According to an article written by The China Post, Acer is hoping to launch their own Intel smartphone before the end of 2012. A potentially odd move if true, especially with CES taking place in early January.

The news comes from Acer's Peter Shieh, during a launch event for the company's new S500 phone. While declining to comment on a release schedule, or any kind of hardware specs, Shieh did indicate that Acer plans to include their own cloud services on the device. If it all comes to pass, it will be another step in the right direction for Intel. It shouldn't come as too much of a surprise either. Acer and Intel are long standing partners in the personal computer space, so a move together into mobile is a pretty logical next step.

Personal Growth Is The Same As Individual Development

Almost anyone you know has the ability to give you insight into who you are from their perspective. Your friends, family and colleagues all know you pretty well.

TIP! Rid your life of unnecessary stress. You only create more stress than before when you over-react to negative events, and that is pointless.

You have to be in good physical shape if you want your personal development efforts to be as effective as possible. Simple activities like getting enough sleep, regular exercise and a healthy diet help your energy level remain high, giving you a greater chance of success on your self improvement quest. While it may sound simple, this is sometimes one of the most difficult things to do.

TIP! Encouraging faith by love will lead to a higher level of personal development. Faith cannot exist without love.

Begin today setting some money aside regularly for emergencies. This can help you to avoid always turning to your credit cards every time something unexpected arises. If you take a few dollars and put them into an emergency fund every week, you will build a nice amount of money to use whenever something unexpected arises. That fund can help us both in the short term and in the long term as our debt decreases instead of grows.

TIP! It helps to see your good points in writing. Write down everything you like about yourself.

Track your progress better through proper organization. Take your major goals, and break them down into small, manageable components. You can then achieve each of these minor goals more quickly. Monitor how you are doing using your daily planner, or you can make a journal that is specific to your goals for self improvement.

TIP! It is possible to handle difficult situations without allowing yourself to become overly emotional. If you can remain calm during stressful situations, you will feel confident enough to face anything.

Accomplish the things in life you want to accomplish. Not taking action to change your life just means you?ll be stuck with your current one forever. Take action on your dreams and do what you can to make them a reality.

TIP! Taking steps to improve your health is a very important part of personal development. In order to maintain a healthy body, you have to stay motivated to practice healthy eating habits and exercise.

Scrutinize your heart, soul and mind rather than being engrossed with your physical appearance. In the end, your looks do not matter that much. It is your personality and character traits that are going to define who you are in life. Work on improving yourself as a whole person rather than superficial changes.

TIP! Make each day a better one than the last. You should always try to improve yourself.

Research positive habits that have helped others become successful. Pick out the most essential success habits from each person you research, then practice these habits until they embed themselves into your daily routine. Persevere in developing good habits, realizing that they come in time. Many people accept the adage that habits are engrained within you in as little as 21 days.

TIP! The first step in any personal development program is the realization that you have room for improvement. Humans are just a very small bit of the universe.

A good attitude is essential to personal growth. A negative attitude won?t help you advance in life and may actually lead you down the wrong path. Instead, let go of negative thinking. Make a choice everyday to look on the positive side of things and watch your goal progression improve.

TIP! In order to expand your horizons, you should first make a list of goals. As an example, if confidence is what you are after, jot it down.

You should feel comfortable in your own skin and being the person that you are. Remember that everyone is unique, and do not feel bad if you are not the best at everything. Don?t worry about the skills you haven?t acquired yet, focus your attention on the skills you are already good at.

TIP! None of us can say that we haven?t made mistakes from time to time. One of the biggest mistakes that we often choose to ignore is our choice in foods.

It is important to know yourself and accept who you are as a person. Treat yourself kindly and be honest with yourself as the years pass, and enjoy knowing the one person who knows you best

Video: Engineered flies spill secret of seizures

Friday, October 12, 2012

In a newly reported set of experiments that show the value of a particularly precise but difficult genetic engineering technique, researchers at Brown University and the University of California?Irvine have created a Drosophila fruit fly model of epilepsy to discern the mechanism by which temperature-dependent seizures happen.

The researchers used a technique called homologous recombination ? a more precise and sophisticated technique than transgenic gene engineering ? to give flies a disease-causing mutation that is a direct analogue of the mutation that leads to febrile epileptic seizures in humans. They observed the temperature-dependent seizures in whole flies and also observed the process in their brains. What they discovered is that the mutation leads to a breakdown in the ability of certain cells that normally inhibit brain overactivity to properly regulate their electrochemical behavior.

In addition to providing insight into the neurology of febrile seizures, said Robert Reenan, professor of biology at Brown and a co-corresponding author of the paper published this week in the Journal of Neuroscience, the study establishes the fruit fly model as a viable genetic platform for the study of epilepsy and validates the use of homologous recombination, which could also be applied to other genetically linked diseases.

"This is the first time anyone has introduced a human disease-causing mutation overtly into the same gene that flies possess," Reenan said.

Engineering seizures

Homologous recombination (HR) starts with the transgenic technique of harnessing a transposable element (jumping gene) to insert a specially mutated gene just anywhere into the fly's DNA, but then goes beyond that to ultimately place the mutated gene into exactly the same position as the natural gene on the X chromosome. HR does this by outfitting the gene to be handled by the cell's own DNA repair mechanisms, essentially tricking the cell into putting the mutant copy into exactly the right place. Reenan's success with the technique allowed him to win a special grant from the National Institutes of Health last year.

The new paper is a result of that grant and Reenan's collaboration with neurobiologist Diane O'Dowd at UC?Irvine. Reenan and undergraduate Jeff Gilligan used HR to insert a mutated version of the para gene in fruit flies that is a direct parallel of the mutation in the human gene SCN1A that causes febrile seizures in people.

When the researchers placed flies in tubes and bathed the tubes in 104-degree F water, the mutant fruit flies had seizures after 20 seconds in which their legs would begin twitching followed by wing flapping, abdominal curling, and an inability to remain standing. After that, they remained motionless for as long as half an hour before recovering. Unaltered flies, meanwhile, exhibited no temperature-dependent seizures.

The researchers also found that seizure susceptibility was dose-dependent. Female flies with mutant strains of both copies of the para gene (females have two copies of the X chromosome) were the most susceptible to seizures. Those in whom only one copy of the gene was a mutant were less likely than those with two to seize, but more likely than the controls.

While the researchers at Brown compared the seizure susceptibility of whole flies, O'Dowd, lead author Lei Sun, and colleagues at Irvine studied individual fly neurons implicated in seizure activity to see how they behaved as the brains were heated. What their measurements revealed in the mutant flies were flaws in how "GABAergic" neurons take in sodium through channels in the cell membrane. Under normal circumstances, the neurons inhibit brain overactivity. But the mutants' mishandling of sodium led them to fail electrically.

"When [O'Dowd's team] isolates those currents due to the sodium channel, which is what's mutated in this case, and she compares the normal animals to the disease-model animals, what happens is the mutant channels pass too much current," Reenan said. "The channels open too easily and they take more effort to close. They open too soon and they close too late. That effect is magnified at higher temperature. Then the neuron can't send any [inhibitory] signals."

Searching for therapies

With a useful genetic model of epilepsy in fruit flies, Reenan said he is optimistic that researchers can now look for potential treatments for the disease. The next step, he said, is to use the practice of "forward genetics" to look for further mutations that might counter febrile seizures.

Given thousands of flies with model of the disease, scientists can purposely subject them to different DNA-altering conditions and then look to see if any flies lose their propensity for seizures. Among those that do, the researchers can then identify the specific genetic alteration responsible and determine whether that could ever be clinically applied. For example, if it turns out that a mutation proves therapeutic because it causes a certain protein to be overexpressed, then perhaps that protein could be refined into some kind of biologic pharmaceutical.

Reenan said he'd expect to see researchers follow a similar roadmap for other diseases as well.

"Knock-in of specific disease-causing mutations into the fly genome has the potential to provide a rapid and low-cost platform for studying the cellular mechanisms of heritable human diseases," the authors wrote. "In addition, knock-in flies can be used in combination with forward genetic screens to identify suppressor and/or enhancer mutations, a strategy that is challenging in humans and rodent models but well established in Drosophila."

Why Holding A Field Radio To My Ear - Business Insider

It was early, a good friend was calling and the phone was ringing.

I hate the phone; anyone who knows me, knows this. It is a strange irrationality of mine, but my level of discomfort turns to panic as each ring passes.

Sometimes I flip a switch inside and pick it up, other times step outside myself, watch it play through to the end ? then take a moment to recover.

I hate the phone because I was a platoon radio operator during the battle of Fallujah, when I was nineteen, and every time somebody called me out there it was a serious fucking emergency.

I had to monitor the net (field communication network) for unit reports on friendly movement so that my platoon did not walk into another?s gun fire.

One time I had told a tank that it would be clear to fire on a building, shortly after I watched a dozen Marines from another platoon take cover behind the same building, out of sight of the tank. The tank?s turret shifted and pointed toward the building.

When there are too many people talking on a radio channel, the net gets tied up and I have to wait for a person to stop talking before I can talk to them. I frantically held down the button to my handset repeating over and over, more panicked and more panicked, ?Cease fire, cease fire, cease fire!? When I let go of the button I could hear the tank power down with a sound like a vacuum cleaner and my handset answered back, ?Roger, cease fire.??

Other times I would need the radio to call for a medical evacuation of friends who had been shot or killed or hit by explosives.

Most days my ear was stuck to my handset for eighteen hours and nothing special but during the times that nothing happened a person could not help but to wonder what the next horrible phone call might be.

When I turn my knob to our battalion channel sometimes the breaking news of the day is a friend from another company has just been killed; or I am sleepy on hour seventeen but keep nodding to the sound of empty radio static that makes a noise like television snow while filled with a cold panic that if I go to sleep, my friends would die because of me.

Sometimes my friend Nate Douglass would call my apartment late at night and I would not pick up. I would want to cry for fear, but did not feel well enough to help someone who needed real help. I would take a moment to recover and carry on with the endless web surfing. He just wanted to talk, so did I, but war is a bitch and we both know it.?

One time I picked up the phone for a number I did not recognize and it was Luis Munoz, our old point man.

He had moved back to Mexico after the service and was calling to tell me about the violence he was witnessing, he said it was worse than Fallujah and he had a child to raise. He had been shot through the leg in Fallujah so bad that he was told he would never walk again.

When we reunited Luis was in physical therapy walking with a cane in his early twenties, by the time he was discharged from the Marines as a wounded warrior he was jogging.

Rich Casares had been hit by an enemy hand grenade in Fallujah, which had damaged one of his eyes. The doctors put an air bubble behind it; I had to write him because he was in a Texas Prison, when he wrote me he would ask for a picture of Fallujah that looked really good so he could have it tattooed across his back.

Paul Johnson has a kid and soon will Donald Blais, they live in Connecticut today and during the battle rushed into a burning house to ferry the bodies of their wounded friends, without being ordered to.

One early morning in my dark apartment I picked up the phone for Nate Douglass who had also been hit by an enemy hand grenade.

We had been best friends in Fallujah.

We talked about our struggles coming home and then we talked about the day he'd been hit by the hand grenade. He would reference the morning and I would retort with my perspective of the same thing.

When we got to the operation he would talk about what he saw inside a house while I would tell him what I saw outside of that house. I realized that the story flowed naturally and that if I had the other members of our platoon who were there that day I was sure that they could reconstruct the story with even more depth.

I told Douglass that night that I had an idea for a documentary that would tell a story of real life heroism and struggle that might answer questions for outsiders and those just returning from their story.
